ABSTRACT

Acinetobacter baumannii (A. Baumannii) is an emerging opportunistic pathogen responsible for hospital-acquired infections, and which now constitutes a sufficiently serious threat to public health to necessitate the development of an effective vaccine. In this study, a recombinant fused protein named OmpK/Omp22 and two individual proteins OmpK and Omp22 were obtained using recombinant expression and Ni-affinity purification. Groups of BALB/c mice were immunized with these proteins and challenged with a clinically isolated strain of A. baumannii. The bacterial load in the blood, pathological changes in the lung tissue and survival rates after challenge were evaluated. Mice immunized with OmpK/Omp22 fused protein provided significantly greater protection against A. baumannii challenge than those immunized with either of the two proteins individually. The results provide novel clues for future design of vaccines against A. baumannii.

ABSTRACT

Objective To summarize the thoracolumbar injuries and treatment in children and adolescents.Methods Since 2000,clinical data,surgical methods,efficacy and mechanism of 177 children and adolescents with thoracolumbar spine injury treated in Honghui Hospital of Xi'an Jiaotong University School of Medicine were analyzed and compared.Treatment was in accordance with easy typing method of Honghui Hospital of Xi'an Jiaotong University School of Medicine.The principles and systematic methods of analysis of such damage was emphasized.Clinical,physiological and psychological effects were observed.Results According to easy typing method of Honghui Hospital of Xi'an Jiaotong University School of Medicine.Ⅰ a-c 77 cases,Ⅱ a 40 cases.Twenty-seven cases of Frankel A were unchanged.There were 28 cases that had been restored to Frankel C in 30 cases of Frankel B.There were 2 cases that had been restored to Frankel D in 30 cases of Frankel B.There were 2 cases that had been restored to Frankel D in 27 cases of Frankel C.There were 25 cases that had been restored to Frankel E in 27 cases of Frankel C.There were 30 cases that had been restored to Frankel E in 30 cases of Frankel D.In 177 patients,imaging,VAS,ODI,SF-36,DPQ,MMSE,Barthel and psychology had achieved significant differences before and after treatment for follow-up of 7-12 months(all P < 0.05).Conclusions Simple type is easy to grasp.It applies to children and adolescents treatment of thoracic and lumbar spine injury to good effect.It is a reasonable treatment strategy.

ABSTRACT

OBJECTIVE@#To explore the synergistic effect of basic fibroblast growth factor (bFGF) and vascular endothelial growth factor (VEGF) on the growth of bone tissue.@*METHODS@#A total of 36 rabbits were randomly divided into 4 groups with 9 rabbits (18 sides of the anterior limb) in each group, including group A,B,C, and D. For all rabbits 1.0 cm bone defects was created in both sides of the radius. These bone defect regions were implanted with corresponding composites: group A with calcium phosphate cement (CPC) only, group B with CPC/bFGF, group C with CPC/VEGF while group D with both bFGF and VEGF. At the 3rd, 6th, and 12th week after the operation, 6 specimens from each group were randomly selected. The effects were partly assessed by X-ray film examination, bone mineral density (BMD) measurements, biomechanical test and histological observation.@*RESULTS@#X-ray showed that at the 12th week the bone defects in group D were completely repaired with CPC generally degraded,whereas bone defects in group B and C were only basically repaired. BMD measurements showed that at the 12th week the BMD of group D was significantly higher than that of group B and C (P < 0.05). Biomechanical testing(at the 12th week) showed that the maximum bending load of group D was significantly higher than that of group B and C (P < 0.05). Histological observation indicated that at the 12th week, woven bone had become mature lamellar bone in group D. At the same time, the normal relation of cortical bone and marrow had resumed, and so had the normal structure of trabecula. However, the recanalization of bone marrow cavity could not be seen in group B and C.@*CONCLUSION@#These 3 kinds of composite: CPC/bFGF, CPC/VEGF and CPC/ bFGF+VEGF can promote the growth of bone tissue and speed up the repair of bone defects. The composite of CPC/bFGF+VEGF is better than the other two composites in promoting the growth of bone tissues, indicating that bFGF and VEGF have a synergistic effect on the growth of bone tissue.

ABSTRACT

OBJECTIVE:To study the characteristics of ceftazidime-resistant?-lactamase produced by Escherichia coli.METHODS:The types of2strains of drug fast?-lactamase produced by Escherichia coli were determined initially by K-B slip diffusion method and ampholine electrophoresis method;The plasmid was extracted by alkaline lysis and the PCR ampli?fication and sequencing were conducted;?-Lactamase was counter-extracted by saturated ammonium sulfate,filtrated by Sephadex G-75gel and purified by DE-52anion exchange chromatography;The molecular weight of which was determined by SDS-PAGE and the enzyme kinetics parameters of?-lactamas were determined by ultraviolet spectrophotometry.RE?SULTS:The2strains produced a super-broad spectrum?-lactamase(CTX-M-1V)with isoionic point at8.7and the molecular weight at29kDa,which can hydrolyze cefotaxim and aztreonam but imipenem and which was sensitive to sulbactam(IC 50 =94nmol/L)and tazobactam(IC 50 =5nmol/L).CONCLUSION:CTX-M-1V is a CTX-M type super-broad spectrum?-lactamase sensitive to suppressants.
